Tenapanor

Tenapanor is a medication used to treat irritable bowel syndrome with constipation (IBS-C). It is a once-daily, non-absorbable small molecule that acts locally in the gut to reduce the absorption of sodium and chloride ions. This reduces the amount of water that is absorbed from the intestines, leading to more frequent and less painful bowel movements. Tenapanor is taken orally, usually in capsule form, and is usually taken with food.

Tenapanor is a medication used to treat two specific conditions:

  • Irritable Bowel Syndrome with Constipation (IBS-C) in Adults: This is its primary approved use. IBS-C is a condition characterized by frequent constipation and abdominal discomfort.
  • Chronic Kidney Disease (CKD) with High Phosphorus Levels: For this use, Tenapanor is used as an add-on therapy in adults on dialysis.

Here's a breakdown of how Tenapanor works for each condition:

IBS-C:

  • Acts by increasing fluid content within the intestines.
  • Targets a specific protein called sodium/hydrogen exchanger 3 (NHE3) located in the small intestine and colon.
  • By inhibiting NHE3, Tenapanor reduces the absorption of sodium back into the bloodstream.
  • This leads to more water being drawn into the intestines, resulting in softer stools and increased bowel movements.

CKD with High Phosphorus:

  • Helps regulate phosphorus levels in the blood.
  • Reduces the absorption of dietary phosphate from food in the intestines.
  • This helps maintain a healthy balance of phosphorus in the body for individuals with CKD whose kidneys are no longer able to effectively remove excess phosphorus.

Administration:

  • Taken orally as tablets, typically twice a day (b.i.d.) with or without food.

Side Effects:

  • Common side effects for IBS-C can include diarrhea, abdominal pain, headache, and nausea.
  • For CKD, potential side effects include diarrhea, urinary tract infections, and low blood pressure.

Important Points:

  • Tenapanor is a prescription medication and should only be taken under a doctor's supervision.
  • It may interact with other medications, so it's important to inform your doctor about all current medications you're taking.
  • Tenapanor's effectiveness can vary depending on the individual and the condition being treated.
